im running bortezomib method development with mobile phase as perchloric acid and methanol:THF, peak shape is broad so any1 cn help me out

Good luck. Sorry can't give you much help, I never worked on this molecule myself but I knew someone who did work on the method. They experience issues with peak shape by a RP-HPLC method but eventually were able to improve the shape, although it was never perfect and had quite a bit of tailing. I believe the molecule hydrolyses quite easily due to it having the boron containing moety?

I do not see a problem with the molecule...

yashpal: the reason that you did not get an answer from any of the highly qualified people on this board who could solve your problem is the fact that you did not provide sufficient information. Mobile phase details are important. So is the solvent used to inject the sample, the amount injected, as well as plenty of other things. If you do not provide such information, you will not get an answer, because it is not possible to give you an answer.
