buffer selection mobile phase

Hi

I'm using phosphate buffer 0,05M and ACN (85:15) but the time retention is about 3 min, the pKa of drug that i'm analyzing is about 8... someone can help me?
how can i select the correct mobile phase? and how the sample prepare can cause interference in the analysis?
anyone know a book/article to indicate?
