need non-structural carbohydrates, sugar profile, and "true protein" of Shitake mushrooms.

What exactly is a "true protein"? I know there are "proteins" and all of them are comprised of amino acid structural scheme with various substituents. Is there a "False Protein" ? or some protein moiety that gives a false signal on protein assay? For the other items, just GOOGLE it.

according to the lit, true protein is protein measured by means that do no t include ninhydrin positive compounds) and ammonia/urea content.

jumpshooter, I looked at your other replies and I appear to be the only person you suggested to google for information. You appeared to be very helpful to others. Perhaps it is because you know so little about what I asked. I tried sending you a private message, but apparently you are shooting from behind a screen.

@ vestelshirley: First, I don't think private messaging is a feature that is active on this board. Second, your original request was devoid of detail and came off as something of a missive, I think.

If you have a problem, folks here are generally willing to help, but a single line request that isn't a complete sentence and appears as if you might be asking someone else to do your homework for you won't get far.

As I read it, you've not articulated a problem with chromatography so much as demanded that someone do a literature search for you.

Even Shitake mushrooms are chineese, I would recommend to check the application database of Showa Denko, GL Science and Tosoh.

For the analysis of complex carbohydrates, I suggest to look for publications by Pauline Rudd, who is the worldwide expert in this field. I know that some of these separations can be done on Waters UPLC Amide columns, but I am not an expert on complex carbohydrates.

Sugar profiles on the other hand are easy. Find many application examples on the Waters website using the BEH Amide column. Or, if you give us a bit more info, I might be able to find what you need.

The problem is, many of the sugars are unusual and my not separate well on HPLC columns. I would suggest you look at GC-derivatization papers on sugars - there are many out there.
