How to check the detector is working?

Hello everyone,

I have setup an old HPLC instrument (was not used for 5-6 months) and am new to it. I wanted to check whether the FL detector is working or not I tried few assay which involve derivatization process, however I was not able to get the peaks. Is there any simple and convenient method available ?
Please let me know.

Hi,

If you want to isolate the detector from the rest of the instrument and take all of the guess-work out of the sample preparation by removing the samples and standards entirely, what would you need in order to produce a (fluorescent) response?

Check the manual on how to do a Raman spectrum of water.

Or, remove the column and do a direct injection of tonic water (contains quinine).

You can dispose of the excess tonic water by mixing with an appropriate quantity of gin :wink:

Yeah, that's what I was getting at...but I didn't want to just say do XXX. I think this is the test we did in college for one of our instrumental analysis classes to determine detector operation.
