Teepol analysis by HPLC

Hi Guys

Long time reader, first time poster, but I figure someone else might have done something like this before.

I'm looking into doing some cleaning validation by HPLC, and something we need to look at is Teepol detergent, in particular the dodecyl benzyl suphonic acid which does the actual detergent duty.

I reckon you should be able to detect this by HPLC, so I've been doing a couple of scouting runs. I'm using an Intersil column, 150x2.1mm at 0.5ml/min and a 60 minute 5-100% ACN gradient. However I'm getting heaps of baseline noise, and I'm not sure that this column is the best for the job.

Does anyone have any experience in the area that they'd care to share?

You probably have an Inertsil column and it sounds like it should be fine for the job you have to do. If you run a blank a couple of times and still see lots of baseline problems (peaks more than a few mAU) then try a fresh mobile phase. If there is something on the column it should come off with the first gradient. You might also try just washing the column with pure acn for a few minutes before running your gradient. Your molecule is sufficiently hydrophobic that you should get good retention. Also, the benzyl group will absorb well for UV around 260 nm or so.
