lcmsms analysis of an antibiotic

Dear all,
I am analyzing an antibiotic compound with lc-ms/ms systems i am having a problem with more stabilization time required for the stabilization of the column, (almost 20 runs) and the response of the compound decreases as the batch proceeds,
I have tried different columns and mobile phases,still not getting satisfactory performance can anyone help me in this issue?
Also the system is performing well as there is no change in response of another compound used to check the system performance.
