hplc on conditioned media followed by MS

Hello, I am not familiar with chromatography at all. I am wondering if the following experiment would be possible and what issues I would have to consider. I am working with cell culture systems and viral infections. I see upregulation of my gene of interest following virus infection and I have reason to believe that this is regulated by a soluble factor released upon infection. I would like to identify this factor. An idea was to fractionate my supernatant from virus infection by HPLC. Then analyze the fractions for induction of my gene of interest. The active fractions I would like to analyze by mass spectrometry in order to determine which proteins are present. Would this be a feasable experiment? What kind of alternatives exist? What kind of technical problems might I encounter?
