The HPLC is a PE 250 Isocratic with uv detector but no chart recorder. It has last been used a year ago with, it appears, a phosphate buffer. I have no experience in the operation of any HPLC but have studied theory. My initial questions and concerns are:
1.) Should I do a blank run or manually check for build-up?
2.) What should I use for a blank run?
3.)Any advise for calibrating it?
I would greatly appreciate any guidance. I just need someone to bounce some questions off of. Please, please, pretty please, could could someone point me toward a good starting point.
