Hello everyone,

I changed the filament of my 5973N, since it was broken, and also cleaned the source. After that, the tune has changed compared to my older tunes. Normally, 219/69 is around 90-120% with an intensity of ~400000. The last EM volts were 1247 together with the following values:

69.00: 311232 100.00% Ratio: 1.18
219.00: 377600 121.32% Ratio: 4.38
502.00: 42128 13.54% Ratio: 9.62

Now the parameters are:

EM Volt: 1529

69.00: 466176 100.00% Ratio: 1.09
219.00: 159488 34.21% Ratio: 3.77
502.00: 17496 3.75% Ratio: 10.59

Air/Water and vacuum are okay, I also checked the cables for the right connection. The middle mass is the most important for my analytes, so it would be really nice if somebody has an idea for this problem.

First of all, you need to specify which tune you are using, and make sure you used the same one both before and after changing the source (otherwise you're not comparing apples to apples).

Secondly, does the 219 peak look okay in the tune report? If they are fine and there are no other problems indicated on the tune report, the instrument should be fine. If you need a higher response from 219 relative to 69 in order to do your analysis... well, that's what target tunes are for! You can use the software to adjust the mass filter settings to get you the ratios you need.

Read the help files on the Tune Wizard and/or Manual Tuning. If you are running a particular method like 8260, Agilent has documents explaining how it all works. For example: https://www.agilent.com/cs/library/supp ... f05011.pdf

Thank you for your fast help! Normally, I tune with atune and it worked for me for the last years. But I tried the target tune as you said, and it helped increase the 219. I hope that the new tune will work well for my analytes. Thank you!
