Im using a Dionex UHPLC for peptide purity testing, and I'm wondering how I purge a line when adding a new eluent to the uhplc.
Basically there is a vial of HPLC grade water line A and I'd like to replace it with 1% TFA 99% UHPC grade water.
But I'm unsure on how to do all of this.
My understanding would be.
Replace bottle
Put blank column on
Turn purge valve to the left
Go to pump settings in Chromeleon
Select purge?
What am I missing? What volume should I typically select? Is volume selectable?
