I have a biological sample that’s currently in 0.1 mM phosphate buffer + 50% ACN with 0.1% formic acid. I want to do an MSTFA derivatization on it before GC-MS.

I know water and acids will interfere with MSTFA, and I’m concerned about the phosphate salts too. My thought was to dry the sample first, but I’m not sure if the salts will still be a problem, or what the best cleanup approach would be.

Has anyone dealt with this? Would something like SpeedVac → ACN rinse → dry again (or maybe a SPE cleanup) be sufficient to remove the phosphate/FA before going into MOX/MSTFA?
