Hello Everyone,

I was wondering if I could get any insight into why the tune on my Agilent Ultivo (AJS ESI source) is consistently failing to pass. Quick background: nitrogen generator went down for a week or so whilst the MS remained on (became contaminated?), and subsequently high mass losses in the tune attempts. I then cleaned the front end and ion injector, as well as fully tested/flushed the calibrant delivery system to ensure calibrant is being delivered. Today, I sonicated the nebuliser. This seems to have brought back the high masses and abundances all seem okay. Now the issue seems to be the peak widths and mass deltas. There also seem to be additional peaks on the lower end of the mass spectrum - again, possible contaminants? Every other parameter seems to be fine (compared to last autotune pass report).

I will try to figure out how I could upload my tune report somewhere if anyone wants to view it (otherwise see below for a section of the data). Other things I have noticed: calibrant came out the needle when I switched to waste, but not when directed to MS. This was checked by me removing the nebuliser from the front chamber and using blue roll to assess consistent calibrant flow. When I put the nebuliser back into the spray chamber, I cannot see any spray.

Apologies for the long post, but I really hope someone can share their insight on this, as the Ultivo itself is still rather new to me and there seemingly many things to iron out! Thank you for any assistance from anyone.
