NEW TO LC-MS, NEED TO KNOW ABOUT BUFFER REQUIREMENTS

Hey everyone,
So, I am trying to develop a protocol for quantifying Imidazolepropionic acid (IPA) in my worms. When I analysed the LCMS run, there was a lot of background against IPA. I used water:acetonitrile as buffer A and amide buffer as B. Should I change the buffers and how do I know which buffer to use?

[1] As a scientist, you should use correct terminology; a water/ acetonitrile mixture is not a buffer solution,

[2] LC/MS requires the use of VOLATILE buffer solutions; think ammonium acetate not sodium acetate,

[3] Research the literature to find a suitable buffer system for your particular analysis; all of the LC/MS manufacturers put out applications notes.

Some more thoughts on this:

What is an amide-buffer? Certainly nothing you want to use as an LCMS eluent.

Acids should be more sensitive in negative mode. Use 0.1% acetic acid in A and B.

Methanol is usually better in LCMS than ACN.

And finally I would like to cite a sentence that someone posted here before:
