Good morning everyone;
I want to use the Folch method in order to separate lipids of milk with the aim to analyze them later for HPLC.
My question now is: what volume or proportion of milk is appropriate to separate the fats from the water in milk matrix?
or
What method is the useful in order to carry out the milk separation?

Precipitate the lipo-proteins with sodium lauryl sulfate (mixed 1:10 in ratio), then centrifuge and collect the supernate (this will contain the free complex lipids--be sure to discard the middle flocculent layer), then lower pH to isolate lipids, and run a preliminary scan at UV-Vis ~320 nm to qualify the presence of lipidacious moieties.

The original Folch method has been modified by others to match the lipid profiles of differing milks.

A quick general Google search of " Folch Milk Lipids " revealed plenty of relevant publications that include the modified Folch procedure depending on the type of milk ( human, bovine etc ).
