its limit solid phase verus large number of volitile compounds will be its problem?

SPME doesn't represent the "real fingerprint" of analytes external to the fiber. In other words the adsorption from liquid (or gas) to the fiber is regulated by the relative partition equilibrium. This means that if you need quantitation, multiple calibration level are needs and also different internal standard must be introduced according to the classes of compound present into the sample.

Another drawback is the need of cryofocusing for early eluted compounds if you need to avoid the use of split injection (loss of sensitivity). The latest point was recently solved by me by real on column injection of fiber into the analytical column.

By this means I am now able to measure on the same run gases as nitrogen or argon and heavy molecules without any focusing device.
