Vitamin A Palmitate Powder Form

I have a problem in extracting Vitamin A palmitate in oral solution, i try saponification process but it didnt work. The Vit. A palmitate is not soluble in methanol, acetonitrile but i succeded in using NaOH solution. Can you help me in finding other method in this situation.

Thanks! but it didnt work also because it is insolube in THF.

Depends on what you mean with "soluble". Curiosity led me to just google: it is somewhat soluble in EtOH, usually it seems to be formulated in oils. If it dissolves in oils it should be reasonably soluble in hydrocarbons.
I also wonder about your inability to solubilize via saponoification, but NaOH did this??

I'll try to look up what we've used in the past for A palmitate when I return from vacation Friday. Memory had it that it was tHF, but you seem to say otherwise. Maybe it's a matter of concentration????Thanks! but it didnt work also because it is insolube in THF.

Maybe your vitamine is coated, try to peel it off with diluted HCl

Is it the plan to analyse it by means of chromatography? And if so, do you have a method for that? If still so, what is the utilized mobile phase?
I would expect that the compound is soluble in the mobile phase

OK, I checked. Initially ee dissolved 0.2g vitamin A palmitate (from Roche) in 100 ml DMF, then diluted down further. HPLC was on RP-8 using H2O-THF.

Vitamin A Powder Form used to be a dispersion in gelatin or acacia.
In my experience, you really need to be very careful during extraction ( using actinic glassware etc., and use antioxidants like BHT ) to ensure the molecules stay intact. I think the BP or EP detailed the precautions for the powder form, and it's important to use a very good grade of IPA.
HPLC was by C8 with 20:1 methanol: water. I recall that Roche had useful analytical advice information freely available, perhaps still available, if you ask nicely.
