I just want to know that what solvents may affect the stability of RI detector and what other parameters should be kept in mind while working with RI Detector?

Premix the mobile phase and pump from one channel; best to bypass any mixing valves and go straight to the pump.

As stated, premix your MP. Second, I've found that ACN based MP's tend to stabilize more quickly than MeOH based MP's, at least in RP applications. Why this is, I'm honestly not sure. Other things: Use a PEEK fitting on the RI inlet (lower thermal conductivity than SS) and at a minimum keep the column insulated if you don't have a column oven. If you do have a column oven, set it at the same temperature or slightly warmer than your RI cell temperature. Other than that, be patient. Purging the cell well then recirculating your MP overnight is a convenient way to come in to a fully equilibrated machine in the morning with a nearly full MP reservoir.
