tips for Protein mass spec

good day all

i am interested in doing some protein mass spec, but to be honest do not know where to start.

at the moment I am trying to familiarize (?) myself with it and I am running a standard of Cytochrome C. Mass Spec part worked pretty well when I infused it directly, got nice multiply charged ion and all.

but now the chromatography..... eish (common South African slang for "O my word")

can anybody give me a little advise on Mobile Phase considerations, Columns, or just general tips (learned the hard way! :wink: )

H2O/ACN 0.1%TFA on C18 column. Depending on the protein size, move toward bigger pore size (i.e. 180/300Angstrom)
