Why there is drop in response?

Dear all,
I am working on liphophilic NCE validation, initially i was facing problem of cayyover;on later that problem was solved rightnow i am facing problem of 10 times drop in the response after completion of 10 to 15 injections in LC-MS/MS the same drop in response i have observed in internal standard;
i am using 3mM ammonium acetate buffer with acn as a mobile phase,with thermo C18 hypurity colum, can any one will guide me to overcome the problem.
