Thanks to all for your comments. I am not testing a protein - it is a new pharmaceutical API and as such I cannot say too much about the compound. We've switched to a 250 mm, 5 um Luna C8(2) column using a mobile phase of 50 % ACN:50 % pH 5.5 Buffer. The chromatography is fairly good (symmetry and USP tailing both 1.0, high plate count). We're just looking at shortening the run time.

I know I do... 'see that? That resolution you could drive a bus through? I did that! :-D'

When that doesn't make me happy any more it will be time to find a new job.
