Sielc, I ran the phosphate method on the 100 last night. I am very impressed by the results.
My prevoius efforts have included ion pair and derivative method but this is by far the simplest to date.
I now have to run on the MS as the UV response from my analyte is poor at 200nm so I need to try and gain sensitivity.
If you could let me have an external e-mail I will send you an address so you can let me have details of the 200 method.

The answers/clarification to your questions, briefly:

1. Concentration of amino acids is about 1.2-1.5 mg/ml
2. Injection volume 5 uL
3. It took around 1 h to equilibrate column from ACN-water-TFA=30/70/0.1 (standard test after packing)
4. Cortisol has nothing to do with our products; we are not offering a cure for every problem. We are not addressing problems of detection, low UV activity of compounds, etc. We do have solutions for better and more controllable retention for ionizable compounds due to dual mode of interactions
5. Nothing sells itself. Period. Plus you have to be aware of the product to want to buy it…On average marketing a column takes 5 years and huge investments, and I won’t even go there.
6. We do not send free columns, but will do enough free to take advantage of our technology.

1.-3.: Ok, I don´t see anything special here, then.
4. Had the distinct impression that you have some RP materials. There are other suggestions I made, with lots of ions.
5. Even if one would agree with you, one would be inclined to think that this holds for all manufacturers.
6. Free only if I ruined it. Some manufacturers are more trusty.

have you tried a Silica column in "HILIC" mode? Without such a comparison your "being impressed" is difficult to assess.
