I generally use a single stock and do serial dilutions. Once I used separate weighed API because the compd is solubility limiting in the stock diluent. Does anyone prep. say 150% conc and use different injection volumes for evaluating linearity from say 150% to 50%....peak response vs column loading? This approach does not appear to be in compliance with ICH guideline. I see some pros and cons with this approach.
