mdyo,

Do you want to validate the chromatographic method or
Do you want to measure some results?
If you want to validate, that would be done best with pure medium without analyt and spiking.
If you want to determine the recovery of your analyte after bacterial transformation, it depends on youe experiment.

Thank you Tom and Alex.

I am actually trying to validate a method I have developed for the analysis of a compound secreted by bacteria. This method is going to be published (I hope!) but I don't have enough idea about the validation criteria. This is why I am worried about what method to use in the recovery measurements.
