it is odd that people do not recommend use of mixture solvent, only use one pure solvent. how can they gurantee complete elute of analytes in sample?
do you guys think LH-20 stationary phase is similar to cyno column?
You can *never* guarantee complete elution in liquid chromatography.
Can anybody have idea about removing attached contaminant from LH-20 column bed because after running samples in LH-20 column, I got my LH-20 coulmn colorized. So I want to remove the color. If anybody has idea about this then please replya s soon as possible.
Thank you so much

This is a very old topic, but I recently started using Sephadex LH-20 therefore I encountered the same problems mentioned in the thread. So far I found exceptionally useful the following book:

Preparative gel chromatography on Sephadex LH-20
Hans Henke
Huthig Verlag, Heidelberg, 1195
ISBN: 3-7785-2428-3

It is an eminently applicative book, therefore one will not find lengthy discussions about the underlying theory of separation modes on LH-20. I think this is because the behaviour of the material is so complex (gel filtration + lipophilic + hydrophilic + partition...) that simply there are not enough studies around to accurately make a theory out of it. However it gives practical rules about setups, solvents, sample types, and elution order of real world samples that will make life definitely easier
