I have a very basic question: While developing an RP-HPLC method by increasing an aqueous phase percent in mobile phase, what solvent should be used to prepare sample? Richer in organic or aqueous phase?

In general, the diluent should be the same as the mobile phase. When that is not possible or practical, more aqueous is better.
