Requesting Guidance on Chromatography Technique Optimisation

Hello Everyone,

I'm requesting advice from the community to help me get better at chromatography. Although I've been using chromatography for some time, I'm running into certain difficulties that I'm hoping you can me with.

In particular, I'm having trouble maximising column efficiency and fixing peak shape problems in my chromatograms. I've attempted a number of strategies, such as modifying the flow rate and experimenting with various mobile phases, but I haven't been successful in getting the desired outcomes.

I would also appreciate any advice on the most recent developments in chromatography technology or best practices that could improve my existing setup. I would be very grateful for any insights you might have if you have used sophisticated methods or tools that have improved your work.

Do the LCR, Chromacademy or some other formal LC training program to get a handle on these issues.
Read the Snyder, Dolan, Kirkland book, Practical HPLC Method Development. Any of these will educate you on the meanings of the terms in the chromatography equation and keep you out of the "dead end" rabbit holes that waste the time of numerous novice chromatographers.

For column efficiency, consider using smaller particle sizes, optimizing flow rates (via van Deemter plots), and maintaining good column care (e.g., regular flushing and using guard columns). For peak shape issues, check injection volume, mobile phase pH, buffer strength, and ensure consistent column temperature. Degassing your mobile phase can also help.
