I am working on amino acid analysis on Dionex IC. Have been able to develop a method (an hour long) to resolve all 21 amino acids and it works very well with my standards.
However, when I tested shake flask samples (with minimal media), I see some amino acids but then some huge peak (big bump) keeps eluting in my chromatography towards the middle of the run. I have been centrifuging and filtering the samples with 0.2-micron filter plates, but this contaminant keeps showing up. We added nuclease to get rid of nucleic acid and also did TCA precipitation to also remove proteins. I was wondering if somebody can help with any suggestion for sample prep that I can use to analyze amino acid in fermentation broth or shake flask study.
