Regarding Method Development in HBSS Buffer

Dear Respected Members,

I have been recentally working with method development for CaCO-2 studies for NCEs, in HBSS buffer, i am facing problems with in HBSS buffer, does any body here have any experience working with HBSS buffer? What kind of steps i should take while working with CaCO-2 samples? I know this sounds pretty simple but i am facing lot of problems with it.

Could you give some background information:
-nature of you problems?analytical or assay set up.
Analytic parameters(detection;LC-MS,LC_UV),
type of column,sample pretreatment.

Assay set up parameters(PH value,used cell line,
cell culture,buffer substance.
