Hi everyone,

I am currently using Analyst software version 1.4.2. I am working in the forensics area. At present we use an Agilent 1100 coupled to a 4000 QTRAP. Our method is dilute and shoot using urine as the target matrix for drugs of abuse. The method is relatively quick but the data processing is extrememly laborious and it can take almost 2 weeks (I know!!) to process a batch of 25 samples. I really really urgently need some help from a forensics lab in a regulatory setting to help me to improve the reporting method. I have contacted Applied biosystems but they are not able to alter their reporting templates easily. We run the QTRAP in mrm mode and once the threshold is reached the third quad operates as a ion trap and we generate spectra at 3 different collision energies and also a composite spectra. If the ion trap does not trigger we have to go back to ion ratio's.

If anybody could recommend a laboratory/person that would be willing to help I would be very very grateful.

Thank you in advance.

Do you primarily do relative abundance ratioing for confirmation?
All standard disclaimers apply. My posts are my opinions only and do not necessarily reflect the policies of my employer.
Hi,
sorry to hear that it takes so long to process your data. I would need some more details where it is slowing down. Normally, Analyst Reporter Templates can be relativ easily adjusted to your needs.
Or, in the quantitation module of Analyst you can design and apply 'queries'.
There is also a ready to use solution called Cliquid 'Forensic & Toxicology' containing methods, libraries and automated reports.

If you want expand the methods already setup or add new methods to this software tool your self you would need the Cliquid version 'Drug Screen and Quant'. see at: http://marketing.appliedbiosystems.com/ ... 07_LANDING

an overview is given at https://products.appliedbiosystems.com/ ... tID=602260

Cheers,

Mary thanks for your reply. We confirm by comparing spectra of a reference standard at 3 different collision energies and also a composite spectra with that of the acquired spectra for the sample under same conditions. For this I need to pull out the EPI's manually and check the fits, reverse fit and purity against the retentions time. I have to manually go back if the trap does not trigger or fits are poor and go back to ion ratio's. This involves much building of reports and printing and is quite time consumming. Could you/anyone give me advice on how I can streamline this. Would it be enough in your opinion in the forensic's area to confirm using ion ratio's only and eliminate the trap/epi?

Many thanks in advance!

Many thanks vulture for your reply,
Could you give me details as to how the templates can be modified in Analyst software. Also could you explain the advantage of the design and apply queries section that you mentioned as I am not sure what you mean sorry!!

Yes Our lab is due to install Cliquid last week but instead I am awaiting users feedback on this software. I hope to visit Applied Biosystems in the next two weeks in the UK to evaluate it. I know it pulls out the EPI automatically and you can generate some templates which give fits and spectra etc. For us manually pullyin this our is very time consuming. I have been told that setting up a run using Cliquid is awkward as you cannot copy and paste and also you need to inject from same vial twice as cannot get information on EPI and peak areas for quantitiation in same report. Have you had any of these problems and how have you overcome them.
Also in the forensic area I posted a query above in relation to Mary Carson's query and I wondered what your thoughts were on the way we are currently doing things.

Many thanks in advance for your help with this query also

Hi Killerwhale,

On how to set up the report template, you can use Analyst Reporter ; I think the latest version would be Analyst Reporter 2.0 which enables you to edit and change the template or even make a new template with your company logo - hassle free in your reporting.

Also, if you have Cliquid Screen and Quantitation ( drug toxicology and forensic ), it will be much easier because you can use the Analyst reporter to make a new report template and then transfer it to Cliquid and you will be able to have your own report template with the necessary quantitions, library search using 3 different CE, purity, reverse purity or fit or whichever way u want ; depending on how you want your report is !!!

I dont understand what you mean by you cant copy and paste in Cliquid for a run ; you can easily do that because Cliquid comes with the option that can allows you to do whatever editing to your sample batch - in excel format and you can copy and paste it in your sample batch.

If you're referring to making a new run ( method ), then you can develop this method from Analyst under a project folder and when you open this project folder, you will be able to see the new method in Cliquid - you dont need to copy and paste ( even though you still can do this via the manual option through Window ).

Hope that I have answered your concerns ; if there is anything, do let me know, k? Happy to help you out !!! Cheers !!!
