Dear All,

What are chromatographic conditions required to analyse drug solubility in oils(like corn, seed etc). What are the solvents required to extract to analyse in reverse phase chromotography.

whta precauition to be taken.

Please suggest.

If you are trying to estabish the solubility of a single drug in an oil there may be no need to use chromatography- add excess drug substance to the oil allow to equilibrate.

filter using Glass fibre filter

Dilute oil with a suitable solvent and assess by UV, compare with blank oil treated in the same way.

Vegetable oils are not that soluble in solvents such as methanol, ACN, or DMF, so not so easy to subilize such samples without going more non-polar.
