Can it be possible to do customs calculation by EMPOWER. I am interested in ONLINE Related substance, Assay & method validation calculation customized as per requirement

With Empower you can do custom calculations, within a sample, but also between samples, in a sample set,....
How to do this, see your manual/help. If there are problems, let us know

Is this the same way we do it by creating customs field applicable for a project.

yes indeed, when you go to the configuration manager, select your project, and make a new custom field, you can choose which type of customfield you want to make (sample, peak, result, ....), which each have a different behavior.
