Hi!

I am trying to resolve a 10 residue peptide on a TSK gel amide 80 column ( 1mm ID) using TFA-acetonitrile solvent system. Can you please suggest an initial method?
I cannot use metal salts as i have it going to the mass spectrometer. My peptide sequence is AEDVGSNKGA.

Any suggestions will be greatly appreciated.

Thanks!

What are you trying to resolve it from?

I am trying to resolve a proteolytic digest of the peptide AEDVGSNKGA. I first need to see a good retention of the intact peptide on the column and then work from there. I need to quantify the reduction in peak area as this peptide undergoes proteolysis.

A quick look on Google for "predicted retention of peptides" found this:

One of the columns mentioned is an amide column under normal-phase conditions.
