I worked with Sephadex LH-20 and MeOH as mobile phase. My sample was an hydrophylic lyophilized plant extract (large amount of tannins). Conzentration of sample: 0.4g/ml in MeOH; Application volume: 2.5ml
The fill level in the colum was about 50cm. Diameter 2,5cm.
After the separation (1,5l mobile phase) still a large amount of substance is remaining in the column (Colour pale orange-red). Even by washing the column with Water and HCl (0,1N) did not get out the substances. What has happened? Is the adsorption between substances and Sephadex so high or is there an other answer?
I did not find yet the desription of this proplem in literature.
