I am trying to adapt the European Pharmacopoeia HPLC method for triterpene glycosides in black cohosh by ELSD. I say "trying" because instrument parameter changes have so far not resulted in system suitability criteria being met. The Varian ELSD settings seem to be completely different from whatever instrument was used to validate the EP method. The lowest standard doesn't meet signal:noise and the highest standard tops out the detector. I have a feeling troubleshooting advice on that would be to keep trying different settings, so that's actually not my main question.

I can get a decent exponential calibration fit but the EP requires a log-log. Can I get Chromeleon to do this for me, or do I have to break out Excel? I assume I will need to anyway to calculate results for multiple peaks with only 1 standard, which seems crazy but maybe less so for this detector.

Thanks in advance for any advice!
