Is a standard needed for quantification (i.e., for determining the quantity of a species in the solution)?

To determine the quantity of an eluted species, you can use a typical online concentration detector after the SEC column and integrate the peak. You need to know the sample’s extinction coefficient if you are using a UV detector or its dn/dc value if you use a dRI detector. These values are often determined by sequence (for protein extinction coefficient or protein dn/dc in aqueous solvent), from the literature (for dn/dc of a protein or polymer in a specific solvent), or by direct measurement of known concentrations.
