Dear Friends,

We have validated method in which sample dilution is 100mg/100ml and further 10ml/100ml (100ppm solution).
Can we change the sample preparation to 25mg/250ml (100 ppm) to avoid pipetting error.
If yes, we have to revalidate the method or not!

There are two issues with reducing sample quantity - the relative uncertainty in sample mass will rise (unless you change your weighing procedure as well) and sample inhomogeneity will become more of a problem. These might cancel any improvement due to omitting the dilution step.
