Pie x r2 x length will give you the volume of the column. If you know the packing density and the volume between the particles you can calculate the netto volume of the packing, the bed. The void volume can be messured by injecting a compound which is not retarded. "Retention" time of this compund will give you the volume from the injector through the tubing plus the "dead" volume in the column.
Hope that helps.
