Under Pressure: An Open Access Approach to Purification SFC

What Does it Cover?

At the Novartis Institutes for Biomedical Research in Cambridge, MA, there has been a shift from localized HPLC purification by medicinal chemists to an Open Access (OA) based purification approach managed by a small team of analytical chemists. Supercritical Fluid Chromatography (SFC) has traditionally fallen outside the scope of this OA based approach due in part to the complexity in running these systems. Purification support for medicinal chemistry and drug discovery groups is demanding in that it strives to achieve productivity and fast sample turn around with cost effectiveness and efficiency, all while supporting a diversity of molecules and projects. SFC offers a powerful tool to balance system standardization and maintenance of a number of purification options to address a broad range of projects.

What will you learn?

As an attendee, you will learn how best to implement an OA environment for use with purification scale SFC for achiral analysis. Shifting achiral SFC to an OA environment presents a number of challenges. However, utilizing SFC introduced important advantages to the purification process while maintaining the ease of support and robustness required for an OA technique.

The OA SFC systems address several critical purification needs that were unmet by reversed phase HPLC systems and improve upon some separations currently used in a reversed phase format. Best practices associated with screening procedures, training protocols, workflow design will be highlighted. A variety of challenging samples are analysed, providing context for analytical OA SFC purification. The results confirm the approach as orthogonal to RPLC, minimized salt formations, and useful for polar solutes. Metrics provide clear evidence of this successful approach.
