In Empower 2, I put the concentration value of my active in the "Value" tab of the component editor and this value already incorportates the purity ie 20mg of active dissolved in 100mls and 5mls of this into 100mls for the working standard. Purity of active is 99.5%..

Value = (20/100) * (5/100) * (99.5/100) *1000 = 9.95mcg/mL.

If I put this value in the Empower 3 and leave the Purity blank will it change the value or leave it as is?
We use some custom field to calculate concentration (along with Value but without Purity) so yes, you can include dilutions/purity/etc in the value itself although I find that way much less informative, as oppose to separate data (for your example):

Value = 20mg
InitialVolume = 100ml
Dilution = 20
Purity = 99.5
