Hello

I'm installing a RI detector (refraction index detector) into my HPLC. How do you choose the best operation temperature? I can use any temperature between 35˚C and 55˚C, or better yet, between 95˚F and 131˚F.
Which parameter should I analyse?

Thank you!
I would generally match the detector temperature close/same as the column temperature, and use the column temperature that suits your separation and matrix. I'd keep the tubing to the detector as short as possible, and cover the tubing with insulating foam, even if that looks dumb !!!

And for lower sensitivity assays, I'd turn on the detector the night before and have mobile phase pumping overnight. And I'd pre-mix mobile phase and NOT use a pumps' mixer.
