I am having sensitivity issues with one of my Sciex 3200 LCMS instruments. Upon inspection I notice that one of the o-rings is missing where the ion source meets the mass spec and that an audible hissing sound is heard from the gas leakage in the ion source.

Is the missing o-ring a potential cause of the decreased sensitivity?

Yes,

If you are leaking gas there then you are loosing flow to either the turbo heaters or the flow around the capillary, so you will not be evaporating your effluent properly.

I think if necessary the o-ring is the same size as the inlet liner o-ring on Agilent GC Split/Splitless injection ports.
