Could anybody, please, suggest how can we increase the sensitivity of HPLC-Ms (ESI) detection of steroid hormones and their derivatives in serum?

We have already optimized the method for adult samples but cannot detect some steroids in child samples. We use LLE, water and methanol with 0,1 formic acid as mobile phases, 5mm x 3 μm column.

Your question is not very specific and lacks information.

Start with the instrumental part. Have you optimized the signal for each analyte by infusion, preferably with a T-piece on the mobile phase?

What concentrations are you trying to detect? Have you checked literature/application notes?
