My samples is starch modified with octenyl succinic acid. I try to establish a method for quantification.
I first hydrolyse octenyl succinic acid from starch by adding 4 NaOH and stirring overnight.
My next aim is to remove NaOH prior to LC-MS quantification to aviode ionisation issues.
his is my idea SPE-cleanup (Strong Anion-Exchange):
- Condition with methanol
- Equlibrate with alkaline Water
- Load sample in 4M NaOH
- Wash with methanol
- Elute with Acetonitril with 1-2% formic acid
Any inputs on cleanup and choice of column?
