Separation and quantification of two different HPMC Grades

Dear all
Could you please suggest a HPLC method for separation and quantification of two HPMC grades from a tablet matrix having molecular weight difference of around 1000000.
The problem is both are co-eluting at same RT.

As I recall HPMC is a polymer and will have the same peaks, thus the RT is the same. The grade of HPMC is dependant on the quantitation of the peaks or the main peak (like PCB's). Rather than HPLC, you may be better off measuring the viscosity over time (like corn starch).

Hi Thanks for your response but in a compressed tablet how you can quantify by measuring viscosity as it also contain other excipients.
the concern is separation and quantification of two different grades of HPMC (having difference in molecular weight of around 1000000) from a Tablet.

I hope my concern is clear now.

Hello.
We use https://www.phenomenex.com/Products/HPLCDetail/BioSep-SEC-S/ column for water soluble polymer (chondrotine sulfate).

Your concern is unfathomable to me. HPMC is an excipient and not worthy of quantitation by HPLC or any other 'expensive' instrument. While HPMC will affect validation of a process so will other variables like compression time and pressure. Thus, HPLC will not help you determine the amount or grade of HPMC (if you are trying to re-generate a drug product).

You may have to take a lot and split it to perform a PPQ (process performance qualification). There are no shortcuts!

Have you tried Size exclusion chromatography?
